Abstract

The rapid transformation of the contemporary biopharmaceutical landscape is characterized by the advent of highly sophisticated, non-traditional therapeutic modalities that challenge legacy regulatory systems. This comprehensive review critically analyzes the international regulatory architectures governing nanomedicines, biosimilars, advanced therapy medicinal products (ATMPs), and digital therapeutics (DTx), while assessing the core cross-cutting mandate of pharmaceutical data integrity. By examining the distinct review pathways of the US Food and Drug Administration (FDA) and the European Medicines Agency (EMA), this article maps out the intricate technological, clinical, and data challenges delaying global market access. Through a precise, structured synthesis of 160 critical points supported by robust peer-reviewed evidence, we highlight the urgent need for adaptive, life-cycle-based global harmonization strategies to ensure patient safety without stifling transformative healthcare innovation.
